Biochem/physiol Actions:
The ZFN (zinc finger nuclease) provided in this kit targets the LEMD3 (LEM domain containing 3) gene. LEM proteins are mainly involved in gene regulation, chromatin arrangement and regulation of transcription factor. LEMD3 (LEM domain containing 3) interaction with Smad1/2 (also referred to as MADH (mothers against decapentaplegic homolog)) and Samd3 suppresses BMP (bone morphogenic protein) and TGF (transforming growth factor)-β signaling, respectively. It also plays an important role in the regulation of the clock protein, BMAL1 (brain and muscle ARNT-like 1). Absence of LEMD3 activity causes Buschke-Ollendorff syndrome and sclerosing bone dysplasia.
General description: CompoZr Knockout ZFNs create targeted double strand breaks at a gene specific target locus. Through the cellular process of Non-Homologous End Joining, this double strand break can result in modification of the DNA sequence and therefore create a functional knockout of the targeted gene. To provide the best chance of creating a functional gene knockout, CompoZr Knockout ZFNs are targeted to the first 2/3 of the coding region for the gene of interest.
Legal Information: All Zinc Finger Nucleases are sold under license from Sangamo Biosciences. For a copy of the Label License provided with purchase of CompoZr ZFNs, please visit the ZFN Label License page.
Legal Information: CompoZr is a registered trademark of Sigma-Aldrich Co. LLC
Physical form: CompoZr Knockout ZFNs are provided as 10 aliquots of mRNA encoding for the ZFNs along with plasmid DNA for both ZFN pairs. Also included in the kit are custom-designed primers, that allow for determination of the rate of mutation and for screening of individual clones that have the desired mutational event, and positive control DNA that has been modified using the CompoZr Knockout ZFN.
